Abstract [en]

The objective of this study was to synthesize dendrimers of larger molecular weight than has previously been done at Polymer Factory AB, Stockholm. The approach was to synthesize an AB2 monomer and with it build dendrimers from multifunctional cores, by divergent growth strategy. The completed dendrimers were to exhibit the properties of matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) calibrants. The synthesis should be cheap, facile and reproducible in large scale. The monomer synthesis was examined in the respect of economic and environmental views. Through a two-step reaction, a glutaric acid functionalized benzylidene protected trimethylolpropane (Glu-TMP-Bz) monomer has successfully been synthesized. This molecule was found to be a suitable candidate for dendrimer synthesis with anhydride chemistry. Dendrimers of first and second generations have been synthesized using Glu-TMP-Bz with cores pentaerythriol (PTT), xylitol (XYT) and dipentaerythriol (DiTME). Glu-TMP-Bz modification of four hydroxyl functionalized fourth generation SpheriCal Aqua™ dendrimers was successfully performed. Molecular weights extended to 41 241 Da, exceeding the previous maximum 30 310 Da. A comparison study with the traditionally used BSA calibrant showed that the dendrimer system has great potential as MALDI-TOF calibrant.
