Abstract [sv]

Den här rapporten behandlar framtagning av en 9-punktskalibreringsstandard för masspektrometrisk analys utifrån dendrimerer samt syntetisering av dendrimerer. Utnyttjande av dendrimerer som ett alternativ till proteiner och peptider för masskalibrering har undersökts och jämförts i projektet. Dendrimerer, som är makromolekylära sfäriska polymerer, kan erbjuda monodispersitet på grund av symmetrin som de har i sin syntetisering. Detta leder till en relativt okomplicerad syntes samt att molekyler kan identifieras med avseende på deras exakta massor tack vare en ”state of the art”-kalibrering. Olika kombinationer av upp till 9 stycken SpheriCal-molekyler gjordes för att ta fram en kalibrant i massområdet 750-7500 Da till MS MALDI-TOF. Detta gjordes på uppdrag av Polymer Factory AB för utveckling av en ny, noggrann och konkurrenskraftig produkt. Resultatet tyder på att kalibranten som tagits fram ger en noggrann kalibrering med tydliga signaler som skulle kunna komplettera den befintliga marknaden av kalibranter för MS MALDI-TOF.

Abstract [en]

The purpose of this report is to develop a nine point calibration standard for mass spectrometric analysis using dendrimers and to study the synthesis of dendrimers. The use of dendrimers as an alternative for proteins and peptides for mass calibration has been examined and compared in the project. Dendrimers are macromolecular polymers that come with monodispersity due to the symmetrical synthesis. They offer a relatively simple synthesis and make identification of masses accurate due to precise calibration. Different combinations of up to 9 SpheriCal-molecules were made to create a calibration standard in the mass range 750-7500 Da for MS MALDI-ToF. This was done as a task from Polymer Factory AB for development of a new, precise and competitive product. The results indicate that the created calibration standard gives a precise calibration with clear signals that would have a complementing share of the MS MALDI-ToF calibration standard market.
