Comparison of the biodegradability of different intestinal filling agents

Abstract [en]

The objectives of this project were to study the environmental aspect of some intestinal filling agents used for contrast imaging, after released into the environment. The factor that was investigated was the biodegradability of the agents. The agents were Movprep (main ingredient is PEG), Omnipaque (main ingredient is Iohexol based on Iodine) and a Filling Agent A (protein rich, further called FAA) that currently was during development. The outline of the project included some steps. Movprep and Omnipaque were stated to be stable along human consumption, hence was a step of digestion only carried out for FAA. To add the aspect of solar radiation in the environment, phototransformation experiments were performed. To get a better view of the biodegradability of the agents, the concentration of organic carbon was determined. The environmental issue of exceeded addition of nitrogen compounds was evaluated by analysing the concentration of nitrogen in nitrate. To determine the composition and decomposition, MALDI-TOF MS was carried out. Differences were found between the samples and depending on how long time they were exposed for solar radiation. FAA showed to be rather biodegradable. Movprep was stable, but not as stable as expected. Omnipaque was very stable in elemental concentrations and quite stable in composition, at least for a short time.

Syftet med det här projektet var att studera miljömässiga aspekter av ett par kontrastlösningar använda för tarmen efter att ha blivit utsläppta i miljön. Den faktor som studerades var den biologiska nedbrytbarheten för kontrastlösningarna. Dem innefattar Movprep (huvudingrediens är PEG), Omnipaque (huvudingredient är Iohexol, jodbaserad) and en Kontrastlösning A (proteinrik, kallas nu FAA) som var under utveckling. Dispositionen av projektet bestod av några steg. Movprep och Omnipaque konstaterades att vara stabila under mänsklig nebrytning, följaktligen utfördes steget med nedbrytning endast för FAA. För att lägga till aspekten med solljus i miljön, utfördes phototransformationsexperiment. För att få en bättre bild av bionedbrytbarheten för lösningarna, så analyserades koncentrationen av organiskt kol. Problemet med förstorade mängder kväveföreningar i miljön utvärderades genom att analysera koncentrationen av kväve i nitrat. För att fastställa sammansättningen och nedbrytningen av lösningarna, så utfördes MALDI-TOF MS. Skillnader påträffades mellan proverna och beroende på hur länge dem hade utsatts för solljus. FAA påträffades vara rätt biologiskt nedbrytbar. Movprep var stabil, men inte så stabil som det förväntades. Omnipaque var mycket stabil i ämneskoncentrationer och ganska stabil i sammansättningen, åtminstone under en kort period.
