Abstract [en]

Sequencing is in a golden age but has yet to decipher of pluriploid genomes and complex samples in an efficient manner. Although several methods are capable of obtaining the information needed, associated trade-offs have limited their spread and rendered them alternatives to conventional sequencing experiments. This thesis focused on developing a method capable of producing synthetic long read data without adding substantial amounts of labour or cost to standard short read sequencing. The report features custom filtering of laboratory inferred artifacts and presents a draft protocol that offers a rigid starting point for an independent library preparation protocol for high throughput barcoding of single molecules.
