Optimization of transfection media for rAAV production in HEK293 cells

Genterapi är behandling av en genetisk sjukdom genom överföring av genetiskt material till en patient, i syfte att erhålla långvarigt uttryck för den överförda genen, med tillräcklig intensitet för att orsaka terapeutiska effekter. Adeno-associerade virus (AAV) är en av de mest urskiljbara vektorerna för genterapi, men trots sin framstående kliniska framgång är vektorproduktion fortfarande en flaskhals.

I detta arbete testades flera aminosyror som tillskott för transienta transfektionsmedier. Experimenten gjordes i liten skala, med volymer på 5 ml, för att screena flera parametrar parallellt. Vi utvärderade variationen på kulturernas livskraft och celltäthet under 72 timmar efter transfektion (hpT) i olika mediasammansättningar. Dessutom analyseras transfektionseffektivitet via GFP - produktion genom flödescytometri och fluorescensmikroskop. Slutligen kvantifierades rAAV - produktionstiter med ELISA.

Resultaten stöder strategin för optimering av övergående transfektion och odlingsmedier som ett sätt att öka utbytet av AVV -partiklar i HEK293 -celler. Vi diskuterar viktiga begränsningar för metodens reproducerbarhet och föreslår nya tillvägagångssätt för att ännu mer förbättra studien i framtida experiment.

Abstract [en]

Gene therapy is the treatment of a genetic disease through the transfer of genetic material to a pa tient, aiming to obtain long-lasting expression of the transferred gene, with enough intensity to cause therapeutic effects. Adeno-associated viruses (AAV) are one of the most distinguishable vectors for gene therapy, but despite of its eminent clinical success, vector production is still a bottleneck.

In this work, several amino acids were tested as supplements for transient transfection media. The experiments were done at small scale, with volumes of 5 mL, to screen multiple parameters in parallel. We evaluated the variation on cultures’ viability and cell-density throughout 72 hours post transfection (hpT) in different media compositions. Moreover, transfection efficiency is analysed via GFP production by flow cytometry and fluorescence microscope. Finally, rAAV production titres were quantified using ELISA.

The results endorse the strategy of optimization of transient transfection and cultivation media as a way to enhance the yield of AVV particles production in HEK293 cells. We discuss important limitations on the method’s reproducibility and propose new approaches to improve even more the study in fu ture experiments.
