Abstract [sv]

Genom den molekylärteknologiska utvecklingen har det blivit möjligt att uppmäta många olika variabler i enkla prover. Denna förskjutning av data konstruktion sätter begränsingar på vilka statistiska metoder som är lämpliga för att särskilja relevanta från icke relevant aspekter I datan. I denna avhandling ägnar jag åt att utforska olika metoder för att finna intresanta biomarkörer i två separata sepsis kohorter där båda kohorterna består av fler uppmäta proteiner I jämförelse med antalet patientprover. Metoder som har studerats i denna avhandling inkluderar uttrycksjämförelse mellan de uppmätta proteinerna, selektiv protein extraktion och metoder för dimmensionreducering genom proteinuttryckfusion. De proteiner som fanns intressanta kom sedan att studras genom en lättare literaturstudie. I slutändan fann jag tre proteiner som skulle kunna vara relevanta för att diagnosera E coli pyelonefrit.

Abstract [en]

In recent decades, the number of measurable parameters in biological samples has skyrocketed. The increase in the amount of measurement available to the number of samples puts constraints on the type of analysis that can be performed and what sort of conclusions can be made from such a data set. In this thesis, I aim to uncover methodological strategies for biomarker discovery in two separate sepsis cohorts for two different pathogens where the number of features measured outnumber the number of samples. Methods used include differential expression analysis, feature selection and feature extraction. The unique proteins found to be of interest were then analyzed in a biological context. In conclusion, I found three proteins that could be potential biomarkers for E coli pyelonephritis.
