Abstract [en]
Knowledge of the proteins found in human blood is of great importance for developing a more personalised health care where all patients receive treatment and diagnosis according to their own chemistry. The purpose of this project was to investigate what protein biomarkers found in blood are analysed in Swedish healthcare as well as the possibilities of detecting the proteins by immunoassay, Mass Spectrometry (MS) and Proximity Extension Assay (PEA). FDA-approved biomarkers presented in the study The Clinical Plasma Proteome (2010) by N. Leigh Anderson was compared with data from the Human Protein Atlas to compile information about the protein’s occurrence in blood, disease connection and possible detection methods. The hospital databases Provtagningsanvisning A-Ö by Karolinska Universitetslaboratoriet and Analysportalen by Labmedicin Skåne was used to investigate the protein’s clinical relevance. A list of selected proteins were tested to be detected by mass spectrometry to confirm the result experimentally and to be able to draw conclusions about optimised sample concentrations. From the information compiled from the databases it could be seen that around 90 % of the studied protein biomarkers also analysed in healthcare can be detected using mass spectrometry. In the experiment, all target proteins were able to be identified although with different precision.
