    Spatial proteome profiling of the compartments of the human cell using an antibody-based approach2017Licentiatavhandling, med artikler (Annet vitenskapelig)
    Abstract [en]

    The human cell is complex, with countless processes ongoing in parallel in specialized compartments, the organelles. Cells can be studied in vitro by using immortalized cell lines that represent cells in vivo to a varying degree. Gene expression varies between cell types and an average cell line expresses around 10,000-12,000 genes, as measured with RNA sequencing. These genes encode the cell’s proteome; the full set of proteins that perform functions in the cell. In paper I we show that RNA sequencing is a necessary tool for studying the proteome of the human cell.

    By studying the proteome, and proteins’ localization in the cell, information can be assembled on how the cell functions. Image-based methods allow for detailed spatial resolution of protein localization as well as enable the study of temporal events. Visualization of a protein can be accomplished by using either a cell line that is transfected to express the protein with a fluorescent tag, or by targeting the protein with an affinity reagent such as an antibody. In paper II we present subcellular data for a majority of the human proteins, showing that there is a high degree of complexity in regard to where proteins localize in the cell.

    Cellular energy is generated in the mitochondria, an important organelle that is also active in many other different functions. Today approximately only a third of the estimated mitochondrial proteome has been validated experimentally, indicating that there is much more to understand with regard to the functions of the mitochondria. In paper III we explore the mitochondrial proteome, based on the results of paper II. We also present a method for sublocalizing proteins to subcompartments that can be performed in a high-throughput manner.

    To conclude, this thesis shows that transcriptomics is a useful tool for proteome-wide subcellular localization, and presents high-resolution spatial distribution data for the human cell with a deeper analysis of the mitochondrial proteome.

    Abstract [en]

    Defining the human mitochondrial proteome is crucial to understand the mitochondria’s diverse functions and role in disease. Here, we present an image-based map of the human mitochondrial proteome containing 1,070 proteins, of which 376 have previously not been reported to localize to the mitochondria. Our analysis shows that approximately 50% (n=493) of the proteins localize to additional cellular compartments, likely contributing to the diverse cellular functions of mitochondria. This map of the mitochondrial proteome, part of the Cell Atlas of the Human Protein Atlas database (www.proteinatlas.org), provides a valuable knowledge resource for studies of mitochondria function, dysfunction and disease.

    Abstract [en]

    Radon dissolved in drinking-water can be transferred into the indoor air and is one of the main transfer pathways for radon. At water purification plants, large quantities of water are treated and there is a risk that radon degasses from the water and enters into the indoor air. Hence, there is a risk for elevated radon levels in the indoor air at these facilities. This study aims to investigate the general impact of water treatment processes on the radon concentration in water and its transfer into the indoor air. Moreover, the risk that radon exposure exceeds the regulatory limits at workplaces was investigated. In total, the results from 39 Swedish water purification plants are included in the study. The methodology includes long-term air measurements with alpha track detectors, and short-term air measurements with AlphaGUARDs. In addition, water samples were collected in order to analyze the radon concentration in the untreated and treated water.

    The results show that several plants experience elevated radon levels in the indoor air and in some cases the exposure could be problematic. Several connections were investigated without finding apparent connections for those cases. For example, the relation between radon concentration in the water and radon level in the indoor air was investigated and the connection between the volume of water treated and the radon level in the indoor air. Calculations with transfer coefficients indicate that the transfer of radon into the indoor air is relatively small. However, there can also be contribution from other radon transfer pathways, such as soil and buildings, which may have an impact on the radon levels in the indoor air.

    Nyckeltal används för att jämföra företag och på så sätt värdera dessa. Aktiekursen kommer att variera över tiden och stundtals nåskyhöga priser liksom djupa bottnar. Denna enorma fluktuation var markant under finanskrisen 2008. Den här rapporten undersöker huruvida värdering av företag med avseende på nyckeltal skiljer sig mellan strax före och strax efter finanskrisen. Detta görs genom att modelera värdet av S&P500-industriföretag med hjälp av multipel linjär regression. Resultatet kan inte med statistisk signifikans avgöra om värderingen med avseende på nyckeltal skiljer sig från före och efter finanskrisen.

    New types of cellular networks are appearing on the market, such as GPRS and UMTS. In addition to this, an increasing number of places are being equipped with wireless LAN extensions to the Internet. Simultaneously, new mobile devices are becoming available that can perform complex computations based on events from various input devices and/or information sources. The combination of extended network connectivity and new mobile devices creates new opportunities for network computing.

    This paper presents a scenario where network enabled applications can interact with mobile devices to create a smart space. The ambition is to create a system where mobile devices can be aware of what resources are present at a specific location. The prototype solution in this paper uses an XML-based protocol and a tuple space architecture as a message passing mechanism.

    Längst ut på piren i Värtahamnen har jag velat skapa Operastaden, en mikrostadsdel som är en egen värld helt uppbyggd kring opera. Hit skulle alla operarelaterade verksamheter flytta, såsom operahögskolan och baletthögskolan. 

    Mellan byggnaderna skulle folk springa i sina scenkläder, dekorer skulle lastas in och ut, och sångarna skulle värma upp sina stämmor på gården. Platsen skulle genomsyras av operaproduktion, och ta intryck av den dramatiska omgivande produktionsmiljön i hamnen. Inåt platsen skulle allt vara smyckat som dekorer, och scenografier och olika miljöer skulle vara uppbyggda för att vara experimentscener, där det blir oklart om vem som är besökare och vem som är skådespelare.

    Ta ett bad poolen i Mallis-miljö, eller picnica vid sjön bland tonerna från Verdi, operans förtrollade värld gör att du inte längre behöver bry dig om vad som är fantasi eller vad som är verklighet. 

    Opera är inte längre begränsat till om du har råd att betala en biljett, eller om du är för rastlös för att sitta i stolen genom alla tre akter.

    Mitt projekt handlar om den slutna gården, skogen och de kringbyggda trähusen. En referens till projektet är den traditionella gården där husen är sammanförda i längor runt en gårdsplan. Gårdens slutna form, där husens dörrar vetter in mot gårdsplanen, ger gott om skydd mot yttre naturliga och övernaturliga krafter samtidigt som alla nödvändiga utrymmen i det dagliga arbetet är lätt att nå. På den här förskolan ska man vistas mycket utomhus, antingen på den gemensamma gåden är barnen kan känna sig trygga och övervakade eller ute i skogen där en mer fri lek kan äga rum. Att ha undervisning utomhus i naturen gör att barnen får prova på olika friluftslivsaktiviteter och grundlägger färdigheter som de har nytta av hela livet. De får också en nära relation till naturen och utvecklar kunskap och trygghet att vistas där. På förskolan ska man känna en stark hemvist. Fasaderna är målade i olika lekfulla färger för att barnen ska känna igen sitt eget hus. 

    Förskolan består av tre åldersanpassade avdelningar där varje avdelning får sina egna hemvister. Planen är rationell och ser likadan ut för alla avdelningarn. Detta föratt barnen ska känna igen sig när de byter avdelning. De minsta barnen har dock hela sin avdelning på markplan för att minska transporter i trappor. På markplan ligger även avdelning två och tres allrum, kapprum och toalett. Varje allrum går att öppna upp mot innergården och kan på så sätt bli en del av den. Här kan de olika avdelningarna mötas. Även köket och varuintagningen ligger på markplan och maten körs ut till de olika avdelningarna på rullvagnar. På övervåningen ligger personalavdelningen, de äldre barnens lek- och vilrum samt allmänna ytor som samling, atelje, rörelse- och sagorum. Dessa rum har varierande takhöjd och går att nå via de två trapphusen i varje entre´.

    This study started off during the spring of 2017 and had the goal of with the aid of a competitive analysis create a new social media strategy for the company Spree. During the last years, digitalization has grown increasingly and has become more integrated in our society. Together with the digitalization, the use of social medias has increased, both for personal use and for business use. For companies, the use of social medias is still relatively new, so a designed and developed social media strategy can be crucial for a company to reach out in the right way and to the right target group.

    This study show that a well-designed social media strategy can be created with the aid of a competitive analysis together with a deeper analysis of the company’s own social medias. The analysis of the company’s own social medias helps to illustrate the current flaws that exists, while the competitive analysis assist in finding relationships that can help the company get an apprehension of what their main focus should be on.

    Examensarbetet är utfört vid Volkswagen Parts Logistics i Nykvarn. Företaget är ett reservdelslager för Volkswagen Groups bilmärken på de svenska, finska, baltiska och norska marknaderna.

    Examensarbetets syfte är att ge förslag som förbättrar produktiviteten och förbättra arbetsförhållandena, med bibehållen eller ökad kvalitet på två avdelningar i lagret: Parts Logistics Pallgods och Parts Logistics Avgåendegods. Förändringsförslagen är, eftersom lagret är nybyggt (2006), fokuserade på hur processerna i sig kan förbättras utan ombyggnationer och investeringar.

    Examensarbetet är utfört genom att aktivt arbeta i processerna på de två avdelningarna för att sedan analysera dessa med hjälp av en litteraturstudie och diskussioner med de anställda. Dessutom har två studiebesök gjorts för att få inspiration till olika arbetssätt: Clas Ohlsons centrallager i Insjön och Møller Logistikk, som motsvarar Volkswagen Parts Logistics på den norska marknaden.

    Inom de tre olika områden där förbättringsmöjligheter identifierades har jag presenterat följande förslag och potentiella effekter.

    Abstract

     

    An Asian option is a path-depending exotic option, which means that either the settlement price or the strike of the option is formed by some aggregation of underlying asset prices during the option lifetime. This thesis will focus on European style Arithmetic Asian options where the settlement price at maturity is formed by the arithmetic average price of the last seven days of the underlying asset.

    For this type of option it does not exist any closed form analytical formula for calculating the theoretical option value. There exist closed form approximation formulas for valuing this kind of option. One such, used in this thesis, approximate the value of an Arithmetic Asian option by conditioning the valuation on the geometric mean price. To evaluate the accuracy in this approximation and to see if it is possible to use the well known Black-Scholes formula for valuing Asian options, this thesis examines the bias between Monte-Carlo simulation pricing and these closed form approximate pricings. The bias examination is done for several different volatility schemes.

    In general the Asian approximation formula works very well for valuing Asian options. For volatility scenarios where there is a drastic volatility shift and the period with higher volatility is before the average period of the option, the Asian approximation formula will underestimate the option value. These underestimates are very significant for OTM options, decreases for ATM options and are small, although significant, for ITM options.

    The Black-Scholes formula will in general overestimate the Asian option value. This is expected since the Black-Scholes formula applies to standard European options which only, implicitly, considers the underlying asset price at maturity of the option as settlement price. This price is in average higher than the Asian option settlement price when the underlying asset price has a positive drift. However, for some volatility scenarios where there is a drastic volatility shift and the period with higher volatility is before the average period of the option, even the Black-Scholes formula will underestimate the option value. As for the Asian approximation formula, these over-and underestimates are very large for OTM options and decreases for ATM and ITM options.

    We propose a new lattice Boltzmann numerical scheme for binary-fluid surface interactions. The new scheme combines the existing binary free energy lattice Boltzmann method [Swift et al., Phys. Rev. E 54 (1996)] and a new wetting boundary condition for diffuse interface methods in order to eliminate spurious variations in the order parameter at solid surfaces. We use a cubic form for the surface free energy density and also take into account the contribution from free energy in the volume when discretizing the wetting boundary condition. This allows us to eliminate the spurious variation in the order parameter seen in previous implementations. With the new scheme a larger range of equilibrium contact angles are possible to reproduce and capillary intrusion can be simulated at higher accuracy at lower resolution.

    Abstract [en]

    Measurements of the viscosity of non-Newtonian fluids and suspensions having a solid volume fraction of about 30% or more is of major interest from an industrial point of view. Cement paste and cement grouts for injection grouting applications, with water to cement ratios typically in the range of 0.4 and 0.6 - 0.8 by weight, are two examples of industrial fluid systems. Few in-line techniques are available on the market that can be used for these fluid systems and under realistic field conditions. The so-called UVP+PD in-line rheometry method combining the Ultrasound Velocity Profiling (UVP) technique with Pressure Difference (PD) measurements is a promising new tool for industrial applications. This paper presents an initial pre-study that aims to demonstrate the feasibility of the UVP+PD method using cement grouts for process monitoring and control of grouting applications under realistic field conditions. The UVP+PD method was tested and found successful for continuous inline measurements of concentrated micro cement-based grouts with water/cement ratios of 0.6 and 0.8. The test set-up consisted of a combination of an experimental " flow loop" and a conventional field grouting rig - UNIGROUT, from Atlas Copco. The rheological properties were determined, directly in-line and the parameters obtained were subsequently compared with off-line measurements using a conventional rotational rheometer.

    Abstract [sv]

    Projektet har utvecklats i interdisciplinärt samarbete mellan Jenny Wiklund och KTH, Akademiska Sjukhuset i Uppsala, Uppsala Universitet, Karolinska Institutet, Kungl. Konsthögskolan, Aarhus Arkitektskole, och curator Jan Åman.

    Projektet har utvecklats skulpturalt vid Kungl. Konsthögskolan. Förstudier har genomförts vid Karolinska Universitetssjukhuset. I finalstudien kombineras studier vid Uppsala Akademiska Sjukhuset, Bild- och Funktionsmedicinskt centrum/Röntgen i tredimensionell visualisering av medicinsk bild med studier i klinisk anatomi vid Uppsala Universitet, för att förenas i skulptural gestaltning och iscensättning.

    R1 Reaktorhallen iscensätts som ett platsspecifikt monumentalverk. Den fysiska installationen i rummet bildar en temporal kappa om utställningskapitlen i avhandlingsutställningen som handlar om kropp och hjärna. Utställningen berättar om en specifik tidpunkt, utan minne och proprioception. Här finns ingen historia och ingen framtid.

    Genom utställningen skapas en förståelse för sambanden mellan teoretiskt tänkande och visuell gestaltning. Det är forskning genom konstnärlig och arkitektonisk precision med referenser till konst, arkitektur, medicin och medicinsk teknik. Referenserna är från verkliga objekt i verkliga rum, specifika verk från specifika konstnärer och genom auskultering vid flertalet expertpresentationer, där människor genom närvaro förmedlat sin kunskap.

    Arkitektur är spatial konstruktion av identitet, här i utställningsform som temporal visuell spatial-estetisk poetik. Formen på framläggningen av avhandlingen är vald efter det huvudsakliga ämnet som utreds, minnet, och dess temporala omskapande av rummet vid varje specifik tidpunkt det framkallas. Det visuella intrycket kommer före alla andra beskrivningar, i logik med den händelse som föregick doktorandprojektets ämne, en minnesförlust, där upplevelsen av rummet kom före språket, kroppen före hjärnan, intränade minnen i form av siffror före en självbiografi.

    Abstract [en]

    In this thesis we will talk about why a two-dimensional quantum system gives rise to a particle statistics which neither obey Fermi-Dirac nor Bose-Einstein statistics, but instead ranges continuously between them. One refers to this as anionic statistics and the particles are known accordingly as anyons. The non-abelian category of anyons does a unitary transformation of the wave function upon particle exchange. This exchange braid each particle's world line in the space time which gives rise to non-abelian statistics with applications in topological quantum computations. The main results of our calculations will be to derive the non-abelian statistics by constructing Majorana fermions and study different mathematical representations of the unitary exchange matrices using a Clifford algebraic structure.

    This Thesis describes applications of standing-waveultrasonic traps for sensitive biomedical analysis. Two majorapproaches have been investigated where functionalizedmicroparticles are employed in bioaffinity assays. In the firstapproach, a longitudinal flow-through capillary ultrasonic trapis used for size selective separation and retention ofdifferently sized microparticles. This device may be used fordetection of particle pairs, which are formed during theinitial stage of microparticle immunoagglutination. Theperformance of the capillary ultrasonic trap for enrichment andcounting of particle pairs is characterized by a model systemof differently sized homogeneous fluorescent microparticles.The selectivity of this detection method relies on thecharacteristics of the force field inside the narrow borecapillary, which is formed by the competition between acousticradiation forces and viscous drag forces from the fluidflow.

    The second approach is an investigation of the potential forsensitive protein quantification by combining ultrasonicenrichment and confocal laser-scanning fluore-scence detection.Here, the design of the ultrasonic trap is tailor-made for theimaging properties of a confocal microscope, resulting inrearrangement and concentration of suspended microparticlesinto single, dense layers that is scanned by a focused laserbeam. The bioaffinity assay employed is based on detecting thetarget molecules via fluorescent tracer antibodies immobilizedon the surface of each single particle.

    The final part of the work presented in this Thesis is athorough investigation of both the biochemical and the physicalproperties that determine the performance and potentialsensitivity of the particle doublet assay. In thisinvestigation, a novel approach is presented for doubletdetection, namely fluorescence-microscopy-based classificationof doublets and singlets by a pattern recognition algorithm.The experimental results are also compared with the resultsfrom flow cytometry analysis. Furthermore, the initial stage ofimmuno-agglutination is theoretically investigated by a modelbased on diffusion-limited agglutination combined with a stericfactor determined by the geometry of the bio-molecules and theamount of specific and non-specific binding that is present inthe particular assay.

    To conclude, the Thesis presents several approaches wherestanding-wave ultrasonic fields may be used for sensitiveparticle-based biomedical analysis. The best prospect for highsensitivity was found for the confocal laser-scanningfluorescence detection system, with a detection limit of theorder of 10-14M. On the other hand, the agglutination-basedassay may give sensitivity of the order of 10-11-10-10M with very simple and inexpensiveequipment.

    In this tutorial review in the thematic series "Acoustofluidics", we discuss the implementation and practice of optical microscopy in acoustofluidic micro-devices. Examples are given from imaging of acoustophoretic manipulation of particles and cells in microfluidic channels, but most of the discussion is applicable to imaging in any lab-on-a-chip device. The discussion includes basic principles of optical microscopy, different microscopy modes and applications, and design criteria for micro-devices compatible with basic, as well as advanced, optical microscopy.

    We describe a novel platform for acoustic sample preparation in microchannels and microplates. The utilized method is based on generating a multitude of acoustic resonances at a set of different frequencies in microstructures, in order to accurately control the migration and positioning of particles and cells suspended in fluid channels and chambers. The actuation frequencies range from 30 kHz to 7 MHz, which are applied simultaneously and/or in sweeps. We present two devices: A closed microfluidic chip designed for pre-alignment, size-based separation, isolation, up-concentration and lysis of cells, and an open multi-well microplate designed for parallel aggregation and positioning of cells. Both devices in the platform are compatible with high-resolution live-cell microscopy, which is used for fluorescence-based optical characterization. Two bioapplications are demonstrated for each of the devices: The first device is used for size-selective cell isolation and lysis for DNA-based diagnostics, and the second device is used for quantifying the heterogeneity in cytotoxic response of natural killer cells interacting with cancer cells.

    This review describes the use of ultrasound for inducing and retaining cell-cell contact in multi-well microplates combined with live-cell fluorescence microscopy. This platform has been used for studying the interaction between natural killer (NK) cells and cancer cells at the level of individual cells. The review includes basic principles of ultrasonic particle manipulation, design criteria when building a multi-well microplate device for this purpose, biocompatibility aspects, and finally, two examples of biological applications: Dynamic imaging of the inhibitory immune synapse, and studies of the heterogeneity in killing dynamics of NK cells interacting with cancer cells.

    Several cell-based biological applications in microfluidic systems require simultaneous high-throughput and individual handling of cells or other bioparticles. Available chip-based tools for contactless manipulation are designed for either high-precision handling of individual particles, or high-throughput handling of ensembles of particles. In order to simultaneously perform both, we have combined two manipulation technologies based on ultrasonic standing waves (USWs) and dielectrophoresis (DEP) in a microfluidic chip. The principle is based on the competition between long-range ultrasonic forces, short-range dielcctrophoretic forces and viscous drag forces from the fluid flow. The ultrasound is coupled into the microchannel resonator by an external transducer with a refractive element placed on top of the chip, thereby allowing transmission light microscopy to continuously monitor the biological process. The DEP manipulation is generated by an electric field between co-planar microelectrodes placed on the bottom surface of the fluid channel. We demonstrate flexible and gentle elementary manipulation functions by the use of USWs and linear or curved DEP deflector elements that can be used in high-throughput biotechnology applications of individual cells.

    Ultrasonic radiation forces can be used for non-intrusive manipulation and concentration of suspended micrometer-sized particles. For bioanalytical purposes, standing-wave ultrasound has long been used for rapid immuno-agglutination of functionalized latex beads. More recently, detection methods based on laser-scanning fluorometry and single-step homogeneous bead-based assays show promise for fast, easy and sensitive biochemical analysis. If such methods are combined with ultrasonic enhancement, detection limits in the femtomolar region are feasible. In this paper, we review the development of standing-wave ultrasonic manipulation for bioanalysis, with special emphasis on miniaturization and ultrasensitive bead-based immunoassays.

    A longitudinal hemispherical standing-wave ultrasonic trap for size-selective separation of microspheres in small-diameter capillaries is described. The trap utilizes the competition between acoustic radiation forces and viscous drag forces on spheres suspended in a liquid inside 20-75-mum-diam fused silica capillaries. Experiments performed on 3.0- and 4.7-mum-diam latex spheres demonstrate the principles of trapping and verify the theoretically calculated size-dependent forces on the spheres. The spheres are detected by the use of laser-induced fluorescence. The goal is to use the trap for separation and ultrahigh-sensitivity detection of trace amounts of proteins and other macromolecules containing two antigenic sites, by binding the target molecule with high specificity to antibody-coated latex spheres.

    A novel fluorescence-microscopy-based image analysis method for classification of singlet and doublet latex particles is demonstrated and applied to a particle-based immunoagglutination assay for quantification of biomolecules in microliter-volume bulk samples. The image analysis method, verified by flow cytometric agglutination analysis, is based on a pattern recognition algorithm employing Gaussian-base-function fitting which allows robust identification and counting of singlets, doublets, and higher agglomerates of fluorescent microparticles. The immunoagglutination assay is experimentally modeled by a biotin-streptavidin interaction, with the goal of both theoretically and experimentally investigating the performance of a general immunoagglutination-based assay. For this purpose a theoretical model of the initial agglutination kinetics, based on particle diffusion combined with a steric factor determined by the level of specific and nonspecific agglutination, was developed. The theoretical model combined with the experimental data can be used to optimize an agglutination-based assay with regard to sensitivity and dynamic range and to estimate the affinity, receptor surface density, molecular and binding site sizes, and level of nonspecific binding that is present in the assay. The experimental results are in good agreement with the theoretical model, indicating the usefulness of the model for immunoagglutination assay optimization.

    We combine ultrasonic trapping and capillary electrophoresis (CE) with the goal to detect ultra-low concentrations of proteins via size-selective separation and enrichment of antibody-coated latex spheres. An 8.5 MHz standing ultrasonic wave is longitudinally coupled into the sub-100-muM diam capillary of the CE system. Competition between acoustic and viscous forces result in inflow separation of mum-diam spheres according to their size. Experiments separating 2.8- and 2.1-mum-diam fluorescent latex particles, which model a protein-specific immunocomplex/free particle mixture, indicate a potential improvement of the concentration limit of detection of 10(4) compared to current CE systems. Theoretical calculations show room for further improvement.

    An ultrasonic particle concentrator based on a standing-wave hemispherical resonator is combined with confocal laser-scanning fluorescence detection. The goal is to perform ultrasensitive biomedical analysis by concentration of biologically active microspheres. The standing-wave resonator consists of a 4 MHz focusing ultrasonic transducer combined with the optically transparent plastic bottom of a disposable 96-well microplate platform. The ultrasonic particle concentrator collects suspended microspheres into dense, single-layer aggregates at well-defined positions in the sample vessel of the microplate, and the fluorescence from the aggregates is detected by the confocal laser-scanning system. The biochemical properties of the system are investigated using a microsphere-based human thyroid stimulating hormone assay.

    Ultrasonic manipulation has emerged as a simple and powerful tool for trapping, aggregation, and separation of cells. During the last decade, an increasing amount of applications in the microscale format has been demonstrated, of which the most important is acoustophoresis (continuous acoustic cell or particle separation). Traditionally, the technology has proven to be suitable for treatment of high-density cell and particle suspensions, where large cell and particle numbers are handled simultaneously. In this chapter, we describe how ultrasound can be combined with microfluidics and microplates for particle and cell manipulation approaching the single-cell level. We demonstrate different cell handling methods with the purpose to select, trap, aggregate, and position individual cells in microdevices based on multifrequency ultrasonic actuation, and we discuss applications of the technology involving immune cell interaction studies.

    Abstract [en]

    Computational fluid dynamics (CFD), extensive experimentalstudies and a one-dimensional model based on two pressurereduction coefficients are used in this thesis to analyse thesuction dynamics of an bent axis axial piston pump. Specialattention is given to cavitation since a great deal of problemsare related to this very important phenomenon.

    In previous experimental investigations, a simpleone-dimensional mode derived from the Navier-Stokes equationshas proven relatively accurate in predicting when thecavitation starts in different types of positive displacementpumps. The one-dimensional model, which uses two pump-specificpressure-reduction coefficients to describe theviscosity-dependent and density-dependent pressure drop insidethe pump, was extended and thoroughly analysed in thiswork.

    An extensive experimental analysis of a bent-axis axialpiston pump showed that the one-dimensional model coulddescribe the behaviour of the pump over a fairly wide range ofoperating conditions. Exceptions are at low viscosities incombination with high rotation speeds where the pump deviatesfrom the behaviour predicted by the one-dimensional model.

    CFD simulations in both two and three dimensions were madeon the same geometry as the axial piston pump used in theexperiments. Due to the rotation of the cylinder block and dueto the piston movement, sliding and deforming grids were usedin the three-dimensional CFD analyse. The two-dimensional CFDsimulations suggested two critical regions inside theinvestigated pump where cavities could start to form, insidethe cylinder during the opening and closing phase onto and fromthe valve plate suction port. The results of thetwo-dimensional CFD simulations must however be treated withsome caution, since the agreement with the experimental resultswas poor. The three-dimensional CFD simulations, which agreesbetter with the experiments, suggested a third critical regioninside the pump where cavities could start to from, the areabehind the moving front wall of the cylinder port. A vortexwith low pressure was generated behind the moving front wall ofthe cylinder during the entire suction phase. Thethree-dimensional CFD simulations offered an explanation forthe pump behaviour in the criticalrange of operatingconditions. When the pump operates under non-criticalconditions the lowest pressure is found inside the cylinderduring the closing phase, but when the pump operates in thecritical range of operating conditions, the lowest pressure isfound in the vortex area behind the moving front wall of thecylinder. Since the one-dimensional model assumes that thecavitation always starts in the same position, such a change inthe cavitation point could be a reasonable explanation to thedeviation.

    In general, it is demonstrated that CFD is a useful tool inpump design, even though the results from such analyses must betreated with some caution.

    This study was regarding the waterjets in the water atomization process. This is because the understanding of the waterjets is not complete and with a greater understanding the production of metal powder could be improved.

    The waterjets were going to be categorized according to their wave function, size and distribution of the droplets and the three regimes that Höganäs had divided up the jets into was also analyzed. The three regimes depend on the jets characteristics and the regimes are the transparent, milky and the droplet jet.

    The purpose was to get a better understanding of the correlation between velocity, temperature, waves, size and distribution of the droplets in a 50 cm long waterjet.

    The method to enhance the understanding of this project was to first do theoretical solution with the help of fluid dynamics. Weber, Reynolds and Ohnesorge number were calculated and evaluated to get a better understanding of the waterjet. Secondly, experiments were conducted where a waterjet with different nozzles and temperatures was filmed with a highspeed camera and the videos were analyzed with the help of a software package called ImageJ.

    The results show the correlation between increasing temperature and decreasing droplet size and a less cohesive waterjet core. The conclusion from the study was that with the help of temperature one can help control the droplet size.

    Intermittent disturbances are common in ECG signals recorded with smart clothing: this is mainly because of displacement of the electrodes over the skin. We evaluated a novel adaptive method for spatio-temporal filtering for heartbeat detection in noisy multi-channel ECGs including short signal interruptions in single channels. Using multi-channel database recordings (12-channel ECGs from 10 healthy subjects), the results showed that multi-channel spatio-temporal filtering outperformed regular independent component analysis. We also recorded seven channels of ECG using a T-shirt with textile electrodes. Ten healthy subjects performed different sequences during a 10-min recording: resting, standing, flexing breast muscles, walking and pushups. Using adaptive multi-channel filtering, the sensitivity and precision was above 97% in nine subjects. Adaptive multi-channel spatio-temporal filtering can be used to detect heartbeats in ECGs with high noise levels. One application is heartbeat detection in noisy ECG recordings obtained by integrated textile electrodes in smart clothing.

    Abstract [en]

    Brain computer interfaces (BCIs) are systems that allow users to interact with devices without relying on the neuromuscular pathways. This interaction is achieved by allowing the system to read the electrical activity of the brain and teaching it to map certain patterns of activation to certain commands. There are many applications for BCIs ranging from controlling prosthetics to gaming, but adapting both the user and the system to one another is a time and resource consuming process. Even more problematic, BCIs tend to only perform well for a single user and only for a limited time. This paper aims to investigate the accuracy of single-subject singlesession BCIs on other subjects and other sessions. To that end three different classifiers, a Support Vector Machine (SVM), Convolutional Neural Network (CNN) and Long Short-Term Memory network (LSTM) are developed and tested on a data set consisting of five subjects, two sessions for a binary classification task. Our results show that training on single-subject single-session data leads to an average cross-subject accuracy of 45-50% and an average cross-session accuracy of 50-55%. We find that there is no statistically significant difference in accuracy depending on the classifier used and discuss factors that affect generalization such as model complexity and good subjects.

    Jag har jobbat med att skapa en miljö som jag kallar ”interaktivt suburban”. En ganska klassiskförortstypologi men med offentliga rum. Jag har ritat fem bostadshus som ligger på en höjd i en fåramellan Tyresö centrum, en skola och ett bostadsområde. Här finns närhet till många sporthallar ochidrottsplatser. Förutom bostäder kommer här finnas projekthallar/eventspace och inrymda/programmerade i husens bottenplan, som jag tänker att skolan, de boende och privatagallerier/företag kan utnyttja. Ett slags centrum för folkbildning, skapande och kultur. Formerna ärgenerella men kan inhysa många funktioner. Bostäderna är inspirerade av retro funkisarkitektur. Jaghar jobbat med gamla rumssamband och förnyat vissa delar av bostaden, framförallt köken ochbadrummen. En del dimensioner har krympt och därför är takhöjden i rummen högre för attkompensera. (2,70) Den öppna planen mot gatan framför husen ramas in av en skogsbeklädd brantner mot skolan och fotbollsplanen. En plan anläggs mot bondevägen så att husen får en anslutningtill gatan. På båda sidor av vägen/husen finns möjlighet att odla, leka och ha fest. Tomten är kraftigtterrasserad åt öst. Här ligger även ett boendeparkeringshus. Målet är att locka Tyresöborna tillbakatill förorten eller att det i alla fall ska finnas attraktiva boenden för dem att flytta tillbaka till, efter attde som unga vuxna kanske bott i innerstan. Förhoppningen blir att eventspacen kommer skapa liv iområdet även på kvällarna. Generatorer för detta bygge har vart punkthus från 70-talet,planlösningar från 40-talet och samtiden när det kommer till offentliga rummet ochmarkbehandlingen. Jag har plockat bland referenser inom svensk 1900-talsarkitektur i processen.Ambitionen är att skapa en dynamik i sovstaden.

    What kind of spaces can be traced from a specific sequence of paintings and how does the transition from image to volume work? The Wing is an attempt to approach mental space and to investigate whether an inner space can containtraces of architectural content, assuming that painting as media can work as a bridge to mental space.Many artists such as Wassily Kandinsky, Mark Rothko, Barnett Newman or Swedish Olle Baertling and Ann Edholm have worked with geometry or abstract painting for various, for example rationalist or spiritual, reasons. Their workshare the characteristic that they have many spatial qualities.Roughly we are talking about abstract, yet actual spaces where the dynamic in the surface planes, the open shapes, color space and the memories, intrude on the private space of the beholder and include her or him both physically andmentally.

    There is a growing recognition that community-based organizations (CBOs) show greater success than other development organizations in promoting empowerment. It has also been acknowledged that information and communication technology (ICT) can be a powerful tool for development. In pursuit of empowerment of women, there is an interest to further study CBOs and ICT, and how they can be combined to take further steps forward. The purpose of this thesis is to investigate how mobile phones can be utilized in women's community-based organizations to promote empowerment, conducted as a case study on a women's CBO in Bangalore, India. The study shows that the main organizational characteristics that promote empowerment in CBOs are the participatory approach and the fostering of sense of community. Mobile phones constitute a valuable tool for enhancing these characteristics. However, the study also shows that the digital gender divide and ICT related risks that women face also have organizational implications that have to be considered by CBOs when utilizing ICT.

    Abstract [en]

    The harsh radiation environment of space is known to cause bit flips in computer memory. The conventional way to combat this is through error detection and correction (EDAC) circuitry, but for low-budget space missions software EDAC can be used. One such mission is the KTH project Miniature Student Satellite (MIST), which aims to send a 3U CubeSat into low-Earth orbit.

    To ensure a high level of data reliability on board MIST, this thesis investigates the performance of different types of EDAC algorithms. First, a prediction of the bit flip susceptibility of DRAM memory in the planned trajectory is made. After that, data reliability models of Hamming and Reed-Solomon (RS) codes are proposed, and their respective running times on the MIST onboard computer are approximated. Finally, the performance of the different codes is discussed with regards to data reliability, memory overhead, and CPU usage.

    The findings of this thesis suggest that using an EDAC algorithm would greatly increase the data reliability. Among the codes investigated, three good candidates are RS(28,24), RS(196,192) and RS(255,251), depending on how much memory overhead can be accepted.

    Stockholm stad har en tydlig formulerad målbild för hur Stockholm skall utvecklas till en hållbar storstad där energianvändning och farliga utsläpp är minimala. Denna målbild framgår i stadens Vision för år 2030, där bland annat ett mål om att staden ska vara fossilbränslefri till år 2050 framgår. Staden har utifrån Vision 2030 satt upp en rad miljömål som bland annat innefattar miljöeffektiva transporter och hållbar energianvändning. Utifrån dessa miljömål har en biogasstrategi utvecklats vars syfte är att bidra till en ökad produktion av biogas i regionen.

    Rapporten kartlägger den rådande biogaskedjan för Stockholm stad, från insamling av matavfall till hantering, biogasutvinning och distribution. De olika delmomenten i biogaskedjan analyseras utifrån energiflödet i den aktuella delprocessen.

    Inom Stockholm stad samlas matavfall in från hushåll och verksamheter. Matavfallet samlas in med biogasdrivna sopbilar som kör matavfallet till SRVs förbehandlingsanläggning i Sofielund. Där förbehandlas matavfallet till en homogeniserad, pumpbar massa som kallas slurry. Slurryn lastas på tankbilar som kör det förbehandlade matavfallet till tillgängliga rötningsanläggningar i regionen.

    Vid rötningsanläggningen placeras slurryn i en rötkammare där biogasutvinningen sker. Uppgradering av rågas sker vanligen i anslutning till rötningsanläggningen. Den vanligaste distributionsformen av fordonsgas är i komprimerad form. Rågasen fylls i stålflaskor och transporteras sedan med lastbil.

    Energiinnehållet i biogasen är direkt kopplad till metanhalten. Metanhalten i den producerade biogasen beror på effektiviteten i rötningsanläggningen samt sammansättningen på slurryn.

    Nettoenergivinsten beräknas till 43 procent av energin som fås vid rötning av 1 ton organiskt hushållsavfall.

    De största energiemissionerna sker på grund av långa transporter mellan förbehandlingsanläggningen och rötningsanläggningen. För en energieffektiv biogasproduktion bör samtliga processteg ske i nära anslutning till varandra så att energianvändning och transporter kan minimeras.

    Energianalysen visar att så som biogaskedjan i Stockholm är konstruerad i dagsläget, är den ej förenlig med stadens miljömål om miljöeffektiva transporter och hållbar energianvändning.

    Abstract [en]

    This thesis describes recombinant proteins that have been generated by rational and combinatorial protein engineering strategies for use in subunit vaccine delivery and tumor targeting.

    In a first series of studies, recombinant methods for incorporating immunogens into an adjuvant formulation, e.g. immunostimulating complexes (iscoms), were evaluated. Protein immunogens, which are not typically immunogenic in themselves, are normally administered with an adjuvant to improve their immunogenicity. To accomplish iscom incorporation of a Toxoplasma gondii surface antigen through hydrophobic interaction, lipids were added either in vivo via E. coli expression, or in vitro via interaction of an introduced hexahistidyl (His6) peptide and a chelating lipid. The possibility of exploiting the strong interaction between biotin and streptavidin was also explored, in order to couple a Neospora caninum surface antigen to iscom matrix, i.e. iscom particles without any antigen. Subsequent analyses confirmed that the immunogens were successfully incorporated into iscoms by the investigated strategies. In addition, immunization of mice with the recombinant Neospora antigen NcSRS2, associated with iscoms through the biotin-streptavidin interaction, induced specific antibodies to native NcSRS2 and reduced clinical symptoms following challenge infection. The systems described in this thesis might offer convenient and efficient methods for incorporating recombinant immunogens into adjuvant formulations that might be considered for the generation of future recombinant subunit vaccines.

    In a second series of studies, Affibody® (affibody) ligands directed to the extracellular domain of human epidermal growth factor receptor 2 (HER2/neu), which is known to be overexpressed in ∼ 20-30% of breast cancers, were isolated by phage display in vitro selection from a combinatorial protein library based on the 58 amino acid residue staphylococcal protein A-derived Z domain. Biosensor analyses demonstrated that one of the variants from the phage selection, denoted His6-ZHER2/neu:4, selectively bound with nanomolar affinity (KD ≈ 50 nM) to the extracellular domain of HER2/neu (HER2-ECD) at a different site than the monoclonal antibody trastuzumab. In order to exploit avidity effects, a bivalent affibody ligand was constructed by head-to-tail dimerization, resulting in a 15.6 kDa affibody ligand, termed His6-(ZHER2/neu:4)2, that was shown to have an improved apparent affinity to HER2-ECD (KD ≈ 3 nM) compared to the monovalent affibody. Moreover, radiolabeled monovalent and bivalent affibody ligands showed specific binding in vitro to native HER2/neu molecules expressed in human cancer cells. Biodistribution studies in mice carrying SKOV-3 xenografted tumors revealed that significant amounts of radioactivity were specifically targeted to the tumors in vivo, and the tumors could easily be visualized with a gamma camera. These results suggest that affibody ligands would be interesting candidates for specific tumor targeting in clinical applications, such as in vivo imaging and radiotherapy.

    We have previously reported strategies for Escherichia coli production of recombinant immunogens fused to hydrophobic peptides or lipid tags to improve their capacity to be incorporated into an adjuvant formulation, e.g., immunostimulating complexes (iscoms). Recently, we also explored the strong interaction between biotin and streptavidin to achieve iscom association of recombinant immunogens. Plasmodium falciparum, Toxoplasma gondii and Neospora caninum antigens have served as model immunogens in the different studies. Generated fusion proteins have been found to be successfully incorporated into iscoms and high-titer antigen-specific antibody responses have been obtained upon immunization of mice. We believe that the different concepts presented, utilizing either hydrophobic peptide or lipid tags, or the recently explored biotin-streptavidin principle, offer convenient methods to achieve efficient adjuvant incorporation of recombinant immunogens.
