Abstract [sv]

Utvecklingen av bärarsystem för mRNA-leverans är avgörande för deras framgång som behandlingar mot sjukdomar. Det finns flera begränsningar med mRNA-läkemedel, bland annat instabilitet, svårigheter att passera cellmembranet och immunogenicitetsproblem som kan lösas med hjälp av lipidnanopartiklar. Lipidnanopartiklar är självmonterade strukturer som bildas genom snabb blandning av en organisk fas som innehåller lipider och en vattenhaltig fas som bildas av bufferten och mRNA (Mashima & Takada, 2022). Lipidsammansättningen i LNP:erna bestämmer deras struktur, storlek och funktionalitet (Eygeris et al., 2022). Därför är det viktigt att ha en etablerad karaktäriseringspipeline. Den normala proceduren omfattar dynamisk ljusspridning för att bestämma storlek och polydispersitetsindex och en RiboGreen-analys för att bestämma inkapslingseffektiviteten (Schober et al., 2024). Ingen av dessa mätningar ger dock information på enkelpartikelnivå, vilket är särskilt viktigt för LNP:er eftersom endast en del av LNP:erna innehåller mRNA och denna fördelning inte kan studeras med grundläggande kvalitetskontroll. I denna avhandling har vi utvecklat en analys för bestämning av antalet kopior av mRNA per lipidnanopartikel. Protokollet för analysen fastställdes och optimerades genom att fastställa koncentrationen av var och en av komponenterna. LNP-koncentrationen sattes till 2E+11 partiklar/mL med 0,005 mol% CY5 PE. SYTO 9-koncentrationen fastställdes till 0,005 mM. Det optimerade protokollet testades på LNP:er som formulerats med olika citratbuffertkoncentrationer. En förskjutning av medianen observerades, den ökade med ökningen av buffertkoncentrationen. Det observerades dock också att SYTO 9 försämrades, så ytterligare replikat är nödvändiga med ny SYTO 9.  Trots detta är analysen optimerad och redo för ytterligare tester. Vi räknar med att analysen kan bli en del av karakteriseringsprocessen för LNP:er där skillnader mellan olika formuleringar kan identifieras och registreras. Ytterligare experiment med in vitro-prestationsanalyser skulle kunna avgöra om skillnaderna är korrelerade med LNP:ernas prestandanivå.  

Abstract [en]

The development of carrier systems for mRNA delivery is crucial for their success as treatments for diseases. There are several limitations with mRNA medicines including instability, challenges to cross the cell membrane and immunogenicity problems that can be solved with the use of lipid nanoparticles. Lipid nanoparticles are self-assembled structures formed by the rapid mixing of an organic phase containing lipids and an aqueous phase formed by the buffer and mRNA (Mashima & Takada, 2022). The lipid composition of the LNPs determines their structure, size and functionality (Eygeris et al., 2022). Therefore, it is important to have an established characterization pipeline. The normal procedure includes dynamic light scattering, to determine the size and polydispersity index and a RiboGreen assay to determine the encapsulation efficiency (Schober et al., 2024). However, neither of these measurements provide information on the single particle level especially important in LNPs as only a part of the LNPs contain mRNA and this distribution cannot be studied with basic quality control. In this thesis, we developed an assay for the for determination of the copy number of mRNA per lipid nanoparticle. The protocol for the assay was established and optimized, fixing the concentration of each of the components. The LNP concentration was set to 2E+11 particles/mL with 0.005 mol% of CY5 PE. SYTO 9 concentration was established at 0.005 mM. The optimized protocol was tested on LNPs formulated with different citrate buffer concentrations. A shift in the median was observed, it increased with the increase of the buffer concentration. However, it was also observed that there was a degradation of  SYTO 9, so further replicates are necessary with new SYTO 9.  Nevertheless, the assay is optimized and ready for further testing. We anticipate the assay could become part of the characterization process of LNPs where differences between formulations could be identified and recorded. Further experiments involving in vitro performance assays could determine if the differences are correlated to the performance level of the LNPs.
