Abstract [sv]

Denna studie fokuserar på genetisk manipulation av bakteriella fabriker för att övervinna problematiken som härrör från naturlig extraktion av förening A. Homolog rekombination och transformation av replikativa plasmider användes för uttryck av heterologa gener i värden för att möjliggöra produktionen. Tidpunkten då substrat tillsattes var en studerad faktor, som påverkar produktionen. Vi kunde uppnå nästan det dubbla i produktionen från den valda organismen vid ett senare tillskott. Dessutom utfördes experimenten för att utvärdera olika geners effektivitet och fastställa den bästa kombinationen. Vi testade två olika villkor och vi uppnådde nästan det dubbla av produktionen i bästa fall. Ytterligare experiment krävs för produktion i industriell skala.

Abstract [en]

This study focus on genetic manipulation of bacterial factories to overcome the problematics derived by the natural extraction of compound A. Homologous recombination and transformation of replicative plasmids were used for the expression of heterologous genes in the host to allow the production. The time at which substrate was added was a studied factor, that has an impact on the production. We were able to achieve almost the double in the production from the chosen organism in case of a later addition.  Moreover, the experiments were performed to evaluate different genes efficacy and establish the best combination. We tested two different conditions and we achieved almost the double of the production in the best case scenario. Further experiments are required for an industrial scale production.
